Source of estrogenic activity in soy is isoflavones. Among soy isoflavones, daidzein and genistein and their glycoside compounds of daidzin and genistin possess estrogenic [4] and anticarcinogenic activities [5, 6], respectively. A study using ovariectomized rats has shown that daidzin and genistin reduce bone loss and bone strength reduction [7], or decrease elevated serum triglyceride levels [8] after an estrogen deficiency. Natural healing or naturopathy is a currently accepted means of improving the quality of life for climacteric or menopausal women. However, such strategies are still classified as an alternative to HRT. Healthy climacteric women need to know through empirical testing whether a natural estrogen mimic could be an effective replacement treatment [9]. Iskflavone is one of the best and widespread potential.
First, we summarize studies that tested soy protein that contained a substantial amount of isoflavones. Because it was recognized that isoflavones could be the bioactive component attributed to soy protein, studies published in the late 1990s and beyond generally stated the amount and type of isoflavones in the soy protein. In 22 randomized trials, isolated soy protein with isoflavones was compared with casein or milk protein, 20, 30 46 wheat protein, 47 or mixed animal proteins.48 50 The range of soy protein was 25 to 135 g d; the range for isoflavones was 40 to 318 mg. LDL or non-HDL cholesterol concentrations decreased in most studies, statistically significantly in 8, with an overall effect of 3% weighted average ; . A recent meta-analysis that included 10 studies published from 1995 to 2002 found a similar percentage reduction in LDL cholesterol with no dose effect.51 Over all studies in Table 1, there is no apparent dose effect; the 8 studies with 50 g of soy protein showed a drop in LDL cholesterol concentration similar to those using a smaller amount of soy, 3% overall Table 1 ; . This cutpoint for daily soy protein intake, 50 g, defines a large amount, half or more of the daily average total protein intake in the United States. No significant effects were evident for HDL cholesterol or triglycerides in most of the studies; the weighted average effects were very small: 1.5% for HDL cholesterol and 5% for triglycerides and ketotifen.
DIETARY INTAKE OF PHYTOESTROGENS Phytoestrogens are found in the seeds, stems, roots or flowers of plants, serving as natural fungicides and acting as part of the plant's defence mechanism against microorganisms. They also are the molecular signals that emanate from the root of leguminous plants that attract specific nitrogen-fixing soil bacteria. The main classes of phytoestrogens are the isoflavones, coumestans and lignans. Isoflavones are receiving a great deal of commercial interest at present; they are found almost exclusively in legumes, the soya bean being the most abundant source. The most important soya isoflavones are genistein and daidzein. Lignans, however, are also an important source of phytoestrogens in the UK diet as they are present in most fibre-rich foods. Of a novel protease-activated receptor 2 pathway. On the basis of these findings, stabilized topical soy preparations containing soybean trypsin inhibitor, Bowman-Birk inhibitor and other soy components were developed. Formulations were screened through both biological and physical tests to ensure that biological activities and stability were retained in the formulation. Chemical analysis indicated that these soy preparations contained a complete spectrum of the soy active components needed for skin care. These formulations retained biological activity throughout a shelf life of 2 y and did not irritate the skin. Further pharmacological and clinical studies demonstrated that these soy preparations provided a wide range of skin care benefits. Applications for these preparations include reducing appearance of pigmentation age spots, melasma and mottled pigmentation ; , reducing ultraviolet-induced erythema and peeling, reducing appearance of acne-related blotches and erythema, delaying appearance of hair regrowth and smoothing and moisturizing the skin. In summary, research into the fundamental properties of soy has led to naturally safe and effective soy preparations that deliver a wide range of valuable skin care benefits. BACKGROUND: Thyroid cancer incidence is three times higher in women than in men. It is one of the five most common types of cancer occurring in young women and in recent Asian immigrants. Female hormones and changeable lifestyle factors e.g., dietary intake ; may be important in thyroid carcinogenesis. METHODS: A population-based casecontrol study of thyroid cancer in women, 20 74 years old, was conducted in the San Francisco Bay area. Of 817 cases, diagnosed in 19951998 19921998 for Asian women ; , and identified through the cancer registry, 608 74% ; were interviewed. Controls were identified through random-digit dialing and frequency-matched to cases on age and ethnicity. Of 793 eligible controls, 558 70% ; were interviewed. Interviews were conducted in six languages. Phytoestrogen consumption was assessed by a food frequency questionnaire and quantified from a recently developed database. RESULTS: All three classes of phytoestrogens examined isoflavones, coumestans and lignans ; and four of the seven specific compounds were associated with a reduced risk of thyroid cancer. The odds ratio for the highest quintile of total phytoestrogen consumption compared with the lowest was 0.64 95% confidence interval: 0.41 0.98 ; . Risk reduction was apparent for both white and Asian women. CONCLUSIONS: Despite the association between soybean consumption and goiter, a major risk factor for thyroid cancer, soy-based foods and phytoestrogens in particular seem to be associated with a reduced risk of thyroid cancer in women. Possible mechanisms include antioxidant effects, direct effects on endogenous estrogen and thyroid hormone.

The evaluation of the public health implications of phytoestrogens is complex. The Working Group on Phytoestrogens of the Committee of Toxicity of the FSA 2003 ; mentions a number of reasons for this complexity: Firstly, epidemiological data suggest that a soy-rich diet is associated with a reduction in the risk of a number of diseases. However, many of these studies are of limited value because they do not address specifically the roles of phytoestrogens. Secondly, it is difficult to interpret experimental results. In experiments, subcutaneous instead of dietary administration is often used and high concentrations of phytoestrogens were given. Furthermore, there are significant interspecies differences in the absorption, distribution, metabolism and excretion ADME ; , making extrapolation of the effects seen in animals to humans complex. In addition, many of the studies were short-term intervention studies in adults that did not address the possibility that exposure to phytoestrogens at an earlier age may influence the risk of disease later in life. Furthermore, a significant proportion of the research has been conducted in populations such as the Japanese and the Chinese and thus, extrapolation to Western populations may be confounded by differences in lifestyle, diet, gut microflora, genetic make-up and ADME. Finally, most research has focused on the isoflavones and thus, relatively little is known about the prenylated flavonoids, coumestans and lignans. The only beneficial effect of phytoestrogens that has been convincingly demonstrated is the effect of isoflavones on osteoporosis. In addition, soy may lower cholesterol levels, but it is not clear whether this effect is due to the isoflavones. The effects on menopausal symptoms, cancer, fertility and the central nervous and immune system are not unequivocal. In general, presently, the studies that lead to the suggested possible association between a high phytoestrogen intake and an increased health lack a dose-response relationship. Therefore, the evidence is generally not strong and a quantitative health benefit assessment is impossible. A proven adverse effect of phytoestrogens on human health is the inhibition of the thyroid function by soy, although this effect is likely not caused by the isoflavones alone. Another possible risk may be the growth-stimulating effect on in vitro breast cancer cells. While this effect cannot be extrapolated to man, it does suggest that there may be possible risks associated with the addition of phytoestrogens to diets at physiologically relevant concentrations Kiely et al., 2003; Kurzer, 2003.

From the FFQ ; and we oversampled subjects who were low or high soy consumers. The blood specimens were collected at the time of interview, i.e. after diagnosis for breast cancer cases. In epidemiological studies of exposure e.g. soy intake ; and disease e.g. breast cancer ; associations, the availability of a biomarker as an independent, objective measure of the exposure of interest is appealing, especially in the case of dietary assessment. As shown in Table III, there is convincing evidence that our FFQ is valid in assessing usual adult intake of soy since blood levels of various soy isoflavones measured in a randomly timed blood draw increased significantly with increasing level of usual adult intake of soy. Importantly, casecontrol status did not significantly influence the association between plasma isoflavone levels and self-reported intake Table III ; , providing evidence that cases and controls were comparable in their recall of soy intake and that possible recent dietary changes in subjects especially among cancer cases ; had minimal impact on the quality of dietary information in the parent casecontrol study. This study gives credibility and strengthens our findings of an inverse relationship between self-reported soy intake and breast cancer risk in AsianAmerican women 4 ; . Among non-soy consumers i.e. quartile 1 of adult soy isoflavone intake ; in this study, the geometric mean blood level of daidzein and genistein combined was 7 nmol l, compatible with results found in other non-soy consumers in studies conducted in the US 14 ; and Finland 15 ; . Among high soy consumers i.e. quartile 4, mean daily isoflavone intake was 26.4 mg ; in this study, the mean blood levels of daidzein and genistein combined 30 nmol l ; and total isoflavones 82 nmol l ; were modest, but not unlike levels that have been reported for subjects with similar self-reported soy isoflavone intake. In a British study, plasma daidzein levels were 5, 8, 39 and 132 nmol l, while plasma genistein levels were 14, 17, 119 and 378 nmol l for women whose respective soy isoflavone intakes were 0.2, 20.0, 17.1 and 48.9 mg day, based on FFQs the corresponding estimated isoflavone intakes based on 7 day food diaries were 0.6, 5.9, 19.3 and 33.7 mg day ; 10 ; . Thus, Asian-American women in this study with a daily intake of 26.4 mg isoflavone displayed blood genistein and daidzein levels that were comparable to plasma levels of British women who consumed ~20 mg isoflavones day. To use genetic models, bioinformatics, and expression profiling to identify the gene responsible for regulating IGF-1 expression and BMD. This work will lead to a clearer understanding of how bone mass is attained in mice. Moreover, because of the homology between mouse and human for this chromosomal region, the findings from this study will have direct public health relevance in respect to identifying a gene that contributes to peak bone acquisition, and could be influenced by environmental factors. Ovarian Tumorigenesis Granulosa cell GC ; carcinoma of the ovarian follicles occur in SWRderived inbred strains of mice during pubertal maturation of the gonad. These spontaneous tumors are heritable, with initiation controlled by an SWR-derived oncogenic allele on distal Chromosome 4, with modifier loci on Chrs 1, 2, 9, and 15, plus a strong QTL on Chromosome X. Frequency ranges from less than 1% to 25% of females at 8 weeks of age, depending on their genetic background. Our genetic analyses of this spontaneous tumor model have continued with an emphasis on the main autosomal and X-linked loci that have a dramatic influence on tumor susceptibility and frequency in young female mice. Studies have shown two linked loci on Chromosome X, Gct4 and Gct6, one or both of which are regulated by parent-of-origin mechanisms. Furthermore, the phenotype of increased tumor frequency associated with Gct4 and Gct6 may be the consequence of gene x gene interaction. The oncogenic locus on mouse Chromosome 4 Gct1 ; is within the homologous chromosome 1p36 region of the human genome that has been associated with different cancers. A concentrated mapping effort is in progress with a unique SWR-derived congenic strain carrying CAST sequences on the distal half of Chromosome 4. Fine mapping of the Gct1 locus, together with gene expression studies of the candidate region, has an excellent chance of providing translational insight to juvenile onset GC tumors in pediatric patients. Tumor frequency can be enhanced by exogenous androgen and suppressed by exogenous estrogen.
